Biological Background: CD85d (LILRB2) Function and Localization

  • Leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2), also known as CD85d, ILT-4, LIR-2, and MIR-10, is an inhibitory receptor primarily expressed on myeloid cells and a member of the leukocyte immunoglobulin-like receptor (LIR) family.
  • Serves as a receptor for class I MHC antigens, recognizing a broad spectrum of HLA-A, HLA-B, HLA-C, HLA-G, and HLA-F alleles, thereby contributing to immune down-regulation and the development of tolerance. Related references: PMID:11169396 PMID:12853576 PMID:16455647
  • Recognizes HLA-G in complex with B2M and a nonamer self-peptide, triggering differentiation of type 1 regulatory T cells and myeloid-derived suppressor cells, both of which actively maintain maternal-fetal tolerance. Related references: PMID:16455647 PMID:20448110 PMID:27859042
  • Competes with CD8A for binding to class I MHC antigens and inhibits FCGR1A-mediated phosphorylation of cellular proteins and mobilization of intracellular calcium ions. Related references: PMID:11875462 PMID:12853576 PMID:9548455
  • Is a single-pass type I membrane protein localized to the cell membrane and contains several immunoglobulin-like domains, disulfide bonds, and glycosylation sites.
  • Expressed in monocytes, myeloid and plasmacytoid dendritic cells, tolerogenic IL-10-producing dendritic cells, myeloid-derived suppressor cells during pregnancy, and at low levels in natural killer cells and B cells.
  • Subject to alternative splicing, alternative promoter usage, and phosphorylation, which may modulate its function and expression.

Experimental Guidance and Technical Tips

  • The immunogen used corresponds to the extracellular domain (aa 22–461) of human CD85d, suggesting the antibody likely recognizes epitopes within this region; consider validating reactivity against native CD85d on relevant cell lines.
  • Since the antibody cross-reacts with CD85e, CD85i, and CD85j, careful experimental design (e.g., blocking with recombinant proteins or using knockout/knockdown models) is recommended to confirm target specificity in flow cytometry.
  • For flow cytometry, titrate the antibody to determine the optimal concentration for your specific sample type and staining conditions; PE conjugation is compatible with common compensation setups when using multicolor panels.
  • The antibody is supplied on an ice bag; upon receipt, store at 2–8°C and protect from light to preserve fluorophore stability.
