Biological Background: CD81 Function and Localization

  • CD81 is a 26 kDa tetraspanin, also known as TAPA-1 or TSPAN28, belonging to the tetraspanin family of cell surface proteins that organize signaling microdomains.
  • In B cells, CD81 facilitates the assembly of CD19 and B cell receptor complexes at tetraspanin-enriched microdomains, lowering the antigen threshold required for clonal expansion and antibody production. Related references: PMID:15161911, PMID:20237408
  • In T cells, CD81 localizes CD3 zeta at immunological synapses with B cells, promoting costimulation and T helper type 2 polarization. Related references: PMID:22307619, PMID:23858057, PMID:8766544
