Biological Background: CD62P/P-selectin Function and Localization
- P-selectin, also known as CD62P, GMP-140, PADGEM, and LECAM3, is a cell adhesion molecule belonging to the selectin family.
- It functions as a Ca²⁺-dependent receptor that binds to carbohydrate ligands, such as sialyl-Lewis X, on neutrophils and monocytes, mediating leukocyte adhesion to activated endothelial cells and platelets.
- P-selectin facilitates the rapid rolling of leukocytes on vascular surfaces during the initial stages of inflammation through interaction with its counter-receptor PSGL-1 (SELPLG).
- Additionally, it mediates cell-cell interactions via binding to integrins alpha-IIb/beta3 (ITGA2B:ITGB3) and alpha-V/beta-3 (ITGAV:ITGB3). Related references: PMID:37184585
- P-selectin is stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells, and upon activation by agonists, it is rapidly transported to the cell surface.
- Its subcellular location is primarily the cell membrane, with translocation upon stimulation.
- Key structural features include EGF-like domains, sushi repeats, a lectin domain, transmembrane regions, and sites for palmitoylation and N-linked glycosylation.
- The predicted molecular weight of P-selectin is approximately 90.8 kDa (UniProt), consistent with the observed 91 kDa.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (amino acids 42-771) of human CD62P/P-selectin, which contains the lectin and EGF-like domains, suggesting reactivity with the extracellular region.
- For flow cytometry, consider titrating the antibody and including appropriate isotype controls to optimize the signal-to-noise ratio for your specific cell system.
- As P-selectin is rapidly mobilized to the cell surface upon activation, stimulation with agonists such as thrombin or histamine may be necessary for detection on platelets or endothelial cells.
- Validate the antibody in the relevant human sample type (e.g., activated platelets, HUVECs) under your experimental conditions.
