Biological Background: CD6 Function and Localization
- CD6 (T-cell differentiation antigen CD6, also known as T12 or TP120) is a type I transmembrane glycoprotein expressed primarily on T cells, NK cells, and a subset of B cells (B-1 cells).
- CD6 functions as a cell adhesion molecule that mediates cell-cell contacts and regulates T-cell responses through interaction with ALCAM/CD166.
Related references: PMID:15048703, PMID:15294938, PMID:16352806 - It contributes to signaling cascades triggered by TCR/CD3 complex activation and acts as a costimulatory molecule, promoting T-cell activation and proliferation.
Related references: PMID:15294938, PMID:16352806, PMID:16914752 - CD6 is involved in the formation and maturation of the immunological synapse.
Related references: PMID:15294938, PMID:16352806 - CD6 also functions as a calcium-dependent pattern recognition receptor that binds both Gram-positive and Gram-negative bacteria via LPS and lipoteichoic acid, leading to MAP kinase activation and pro-inflammatory cytokine secretion.
Related references: PMID:17601777 - It is localized to the cell membrane and can be shed as a soluble form detected in serum, suggesting roles in both membrane-proximal and systemic immune regulation.
- Tissue expression profile: high in thymus, spleen, lymph node, appendix, and peripheral blood leukocytes; detected on thymocytes, mature T cells, NK cells, B-1 cells, and some brain cells.
- Alternative splicing produces multiple isoforms with molecular weights ranging from 58 to 71 kDa; the protein undergoes N-glycosylation, phosphorylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 18-364 of human CD6) covers the extracellular region containing the ligand-binding domains; this design may favor recognition of native surface CD6 in flow cytometry.
- For flow cytometry, titrate the antibody to determine optimal concentration for your specific cell type; include appropriate isotype control antibodies.
- CD6 is shed from the cell surface; the presence of soluble CD6 in serum or culture supernatants could potentially compete with membrane-bound CD6 staining; consider this in experimental design.
- When analyzing T-cell subsets, note that CD6 expression levels may vary with activation state; validate staining patterns in the relevant sample system.
