Biological Background: CD44 Function and Localization
- CD44 is a cell-surface receptor encoded by the CD44 gene (also known as LHR, MDU2, MDU3, MIC4) in humans, belonging to a family of transmembrane glycoproteins. Multiple isoforms are generated by alternative splicing, with the epithelial isoform (isoform 10) commonly expressed in carcinomas.
- CD44 functions as a receptor that mediates cell-cell interactions, cell adhesion, and migration, enabling cells to sense and respond to changes in the tissue microenvironment (PubMed:16541107, PubMed:19703720, PubMed:22726066).

PMID:22726066 - It engages extracellular matrix components such as hyaluronan, collagen, growth factors, cytokines, and proteases through its ectodomain, and assembles signaling complexes via its cytoplasmic domain, recruiting effectors like PKN2, RhoGTPases, and phospholipase C to coordinate calcium mobilization and actin cytoskeleton reorganization (PubMed:18757307, PubMed:23589287, PubMed:15123640).

PMID:15123640 - CD44 plays a key role in the activation, recirculation, and homing of T-lymphocytes, as well as in hematopoiesis, inflammation, and response to bacterial infection (PubMed:7528188).

PMID:7528188 - Interaction with LGALS9 ligand activates downstream signaling through LCK, ERK, and MAPK pathways, promoting NK cell activation (PubMed:37006235).
Related references:
PMID:37006235 - The protein is predominantly localized to the cell membrane and cell projections such as microvilli; a soluble secreted form is also detected in biological fluids.
- CD44 is expressed in various tissues, including fibroblasts, urine, and placenta; the epithelial isoform is highly expressed in carcinomas but repressed in neuroblastoma (PubMed:25326458, PubMed:36213313, PubMed:37453717, PubMed:32337544).

PMID:25326458

PMID:37453717
PMID:32337544 - The protein undergoes extensive post-translational modifications, including phosphorylation and glycosylation, and functions as a proteoglycan; it also serves as a receptor for foot-and-mouth disease virus internalization.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region (amino acids 20–178) of human CD44, which is suitable for detecting membrane-bound CD44 in flow cytometry. Because the antibody is directly conjugated to PE, it does not require a secondary staining step; optimize the antibody dilution for your specific cell type to minimize background.
- For flow cytometry (FC), use a relevant human cell line or primary cells known to express CD44, and include appropriate isotype controls to account for non-specific binding.
- Since CD44 expression can vary between cell types and activation states, consider including positive and negative controls to validate staining specificity.
