Biological Background: CD43 Function and Localization
- Leukosialin (CD43, sialophorin, GPL115) is the major cell surface sialoprotein of leukocytes, encoded by the SPN gene. It is a heavily glycosylated type I transmembrane protein.
- CD43 regulates multiple T-cell functions including activation, proliferation, differentiation, trafficking and migration. It positively regulates T-cell trafficking to lymph nodes via association with ERM proteins (ezrin, radixin, moesin) (By similarity).
- CD43 biases T-cell differentiation towards a Th1 lineage by negatively regulating Th2 cell differentiation and promoting IFN-gamma expression during TCR activation of naive cells (PubMed:18036228). Related references: PMID:18036228
- CD43 prepares T-cells for cytokine sensing and effector differentiation by inducing expression of cytokine receptors IFNGR and IL4R, promoting their signaling, and mediating clustering of IFNGR with TCR (PubMed:24328034). Related references: PMID:24328034
- Acts as a major E-selectin ligand on Th17 cells, mediating their rolling on activated vasculature and recruitment during inflammation, but not on Th1 cells. Also acts as a T-cell counter-receptor for SIGLEC1 (By similarity).
- Protects cells from apoptotic signals, promoting cell survival (By similarity).
- Subcellular localization includes the plasma membrane, microvilli, uropodium, and can also localize to the nucleus in PML bodies. It is expressed on thymocytes, T-lymphocytes, neutrophils, plasma cells, and myelomas.
- Post-translational modifications: heavily O-glycosylated and sialylated; also phosphorylated and ubiquitinated.
