Biological Background: CD40 Function and Localization
- CD40, also known as tumor necrosis factor receptor superfamily member 5 (TNFRSF5), is a cell surface receptor for CD40LG/TNFSF5. It transduces TRAF6- and MAP3K8-mediated signals that activate ERK in macrophages and B cells, leading to induction of immunoglobulin secretion. Related references: PMID:31331973
- Alternative splicing produces a secreted isoform, in addition to the canonical membrane-bound receptor.
- Localizes primarily to the cell membrane but can also be secreted. The mature protein has a molecular weight of approximately 31 kDa (calculated 30.6 kDa).
- Contains characteristic cysteine-rich repeats, disulfide bonds, and N-linked glycosylation sites, which are critical for ligand binding and receptor stability.
- Expressed on B-cells and in primary carcinomas, reflecting its dual role in adaptive immunity and tumor biology.
- CD40 signaling is essential for B-cell maturation, class switching, and germinal center formation; its dysregulation is implicated in autoimmune diseases and cancer.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 21–193); thus, the antibody is suitable for detecting cell surface CD40 by flow cytometry without permeabilization.
- Consider titrating the antibody in the relevant cell system to determine the optimal concentration that balances signal-to-noise ratio.
- Include appropriate controls such as isotype-matched PE-conjugated antibody and CD40-negative cells to confirm specificity.
- Protect the PE conjugate from prolonged exposure to light and avoid repeated freeze-thaw cycles to maintain fluorescence intensity.
