Biological Background: CD39/ENTPD1 Function and Localization

  • Ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1, also known as CD39, NTPDase1, ATPDase) is a membrane-bound enzyme belonging to the ENTPDase family.
  • Catalyzes the sequential hydrolysis of nucleoside triphosphates (NTPs) to nucleoside monophosphates (NMPs) via nucleoside diphosphate (NDP) intermediates, playing a critical role in regulating extracellular nucleotide levels. Related references: PMID:8529670, PMID:8626624, PMID:8955160
  • By hydrolyzing proinflammatory ATP and platelet-activating ADP to AMP, CD39 blocks platelet aggregation and supports blood flow. Related references: PMID:8955160, PMID:8996251
  • Subcellularly localized to the plasma membrane and enriched in caveolae, consistent with its role as an ectoenzyme.
  • Expressed primarily on activated lymphoid cells, and also in endothelial tissues; highly expressed in placenta, lung, skeletal muscle, and kidney. Related references: PMID:7930580, PMID:8955160
  • Post-translational modifications include glycosylation, palmitoylation, and disulfide bond formation, as indicated by UniProt keywords Lipoprotein, Glycoprotein, Palmitate, and Disulfide bond.
  • Disease associations: missense mutations in ENTPD1 are linked to autosomal recessive hereditary spastic paraplegia (SPG64).
  • Alternative splicing generates multiple isoforms; the canonical sequence has a predicted molecular weight of ~58 kDa, with additional isoforms observed between 34–59 kDa.

Experimental Guidance and Technical Tips

  • The immunogen (aa 255–442) corresponds to an extracellular domain, targeting an epitope accessible on live cells, supporting flow cytometry applications.
  • PE conjugation enables direct detection; consider titrating the antibody to determine the optimal concentration for your cell type and experimental setup.
  • Multiple bands ranging from 34–59 kDa may be detected in Western blot due to isoforms and glycosylation; validate band specificity using appropriate controls in your system.
  • Cross-reactivity is reported for human; verify reactivity if other species are used.
  • For FC, include appropriate isotype controls and viability dyes to ensure accurate gating of CD39-positive populations.
