Biological Background: SLAMF6 Function and Localization
- SLAMF6 is a self-ligand receptor of the signaling lymphocytic activation molecule (SLAM) family, modulating activation and differentiation of various immune cells. Related references: PMID:11489943 PMID:16920955
- In natural killer (NK) cells, it triggers cytolytic activity when expressed at high densities of natural cytotoxicity receptors. Related references: PMID:11489943 PMID:16920955
- Positive signaling in NK cells involves phosphorylation of VAV1 and depends on the adapter SH2D1B/EAT-2. Related references: PMID:16920955
- SLAMF6 promotes T-cell differentiation into a Th17 phenotype, enhancing IL-17 secretion; this costimulatory activity requires SH2D1A/SAP. Related references: PMID:16920955 PMID:22184727
- It facilitates recruitment of RORC to the IL-17 promoter. Related references: PMID:22989874
- SLAMF6 also may act as a negative regulator of humoral immunity, inhibiting T-cell:B-cell adhesion and germinal center formation (By similarity).
- The protein is expressed on the surface of resting and activated NK cells, T- and B-lymphocytes, with increased surface expression on T-cells from systemic lupus erythematosus patients. Related references: PMID:11489943 PMID:22184727
- Subcellular localization is the cell membrane; it is a single-pass type I membrane protein with an immunoglobulin domain.
Experimental Guidance and Technical Tips
- As a PE-conjugated antibody, protect from light and store according to manufacturer recommendations to prevent fluorophore degradation.
- For flow cytometry, titrate the antibody to determine the optimal working concentration for your cell population and staining conditions.
- The immunogen corresponds to the extracellular domain (aa 24-215), ensuring recognition of the native cell surface receptor.
- The predicted molecular weight is 37 kDa, but due to glycosylation and alternative splicing, bands between 24 and 37 kDa may be observed in non-FC applications like Western blot; validate accordingly.
