Biological Background: CD300a/c Function and Localization
- CD300a, also designated CMRF35-like molecule 8, CMRF35-H9, IRp60, or IgSF12, is an inhibitory receptor of the CD300 family of immunoglobulin-like proteins.
- It functions as an inhibitory receptor that down-regulates cytolytic activity in natural killer (NK) cells and mast cell degranulation. Related references: PMID:10746781 PMID:16339535 PMID:9701027
- CD300a negatively regulates Toll-like receptor (TLR) signaling mediated by MYD88, but not TRIF, through activation of the phosphatase PTPN6. Related references: PMID:22043923
- The protein is broadly expressed on immune cells including NK cells, T-cell subsets, B-cells, dendritic cells, mast cells, granulocytes, and monocytes.
- Subcellularly, CD300a localizes to the cell membrane as a single-pass type I transmembrane protein containing an extracellular immunoglobulin-like domain.
- Key structural features include disulfide bonds, N-linked glycosylation, and phosphorylation sites, consistent with its role in cell adhesion and immune regulation.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (amino acids 18–178) of CD300a, sharing homology with CD300c; this PE-conjugated antibody is designed for flow cytometry (FC) to detect cell surface CD300a/c on live human immune cells.
- For FC applications, perform antibody titration to determine the optimal working concentration for your specific cell type and experimental system.
- Use appropriate isotype controls and viability dyes to ensure specific staining and exclude dead cells.
- Because CD300a is expressed on multiple leukocyte subsets, consider co-staining with lineage markers (e.g., CD3, CD19, CD56) for population-specific analysis.
- Validation in the relevant sample system (e.g., PBMCs, purified NK cells) is recommended to confirm reactivity and performance.
