Biological Background: CD30 Function and Localization
- CD30, also designated TNFRSF8, is a member of the tumor necrosis factor receptor superfamily and is known as Ki-1 antigen and lymphocyte activation antigen.
- It acts as a receptor for TNFSF8/CD30L (PubMed:8391931) and may regulate cellular growth and transformation of activated lymphoblasts. Related references: PMID:8391931
- CD30 activates NF-kappa-B signaling, thereby modulating gene expression (PubMed:8999898). Related references: PMID:8999898
- The gene encodes multiple protein isoforms via alternative initiation and alternative splicing.
- CD30 localizes to the cell membrane and cytoplasm, consistent with its function as a transmembrane receptor.
- At the protein level, CD30 has been detected in alveolar macrophages.
- Post-translational modifications include disulfide bond formation, glycosylation, and phosphorylation; the protein contains a signal peptide, extracellular repeat domains, a transmembrane helix, and a cytoplasmic tail.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 19-379) corresponds to the extracellular region of CD30, making this PE-conjugated antibody suitable for surface staining of live cells in flow cytometry.
- Consider testing the antibody on activated lymphocytes or alveolar macrophages, where CD30 expression has been reported.
- For intracellular detection, a permeabilization step is recommended due to the cytoplasmic localization of CD30.
- Optimal concentration and staining conditions should be determined by the end user.
