Biological Background: TLR2 Function and Localization
- TLR2 (Toll-like receptor 2, also known as TIL4 or CD282) is a key pattern recognition receptor that mediates innate immune responses to bacterial lipoproteins and other microbial cell wall components. It cooperates with LY96 and uses MYD88 and TRAF6 adaptors to trigger NF-kappa-B activation and pro-inflammatory cytokine secretion. Related references: PMID:10426995, PMID:10426996
- TLR2 forms functional heterodimers with TLR1 or TLR6 to discriminate between triacylated and diacylated lipopeptides, respectively, and recognizes mycoplasmal MALP-2, bacterial soluble factors, and Borrelia OspA lipoprotein. Related references: PMID:11441107
- The receptor localizes to the cell membrane, cytoplasmic vesicles, and phagosome membranes, and is enriched in lipid raft microdomains for signaling complex assembly.
- TLR2 is highly expressed in peripheral blood leukocytes, particularly monocytes, and in bone marrow, lymph node, and spleen, with lower levels in lung and fetal liver.
