Biological Background: ICOS ligand Function and Localization
-
Functions as a ligand for the T-cell-specific surface receptor ICOS, delivering a costimulatory signal essential for T-cell proliferation and cytokine secretion.

PMID:30498080 -
Induces B-cell proliferation and differentiation into plasma cells, contributing to humoral immunity.
-
May play an important role in local tissue responses to inflammatory conditions and in modulating secondary immune responses by co-stimulating memory T-cell function (By similarity).
-
In endothelial cells, required for proper neutrophil transmigration in response to chemoattractants such as CXCL8/IL8 and fMLP.

PMID:30498080 -
Localizes to the cell membrane as a type I transmembrane protein.
-
Expressed on peripheral blood B-cells and monocytes, monocyte-derived dendritic cells, and widely in lymphoid and non-lymphoid tissues including lymph nodes, spleen, leukocytes, and on activated monocytes/dendritic cells.
-
Predicted structural features include an immunoglobulin domain, disulfide bonds, N-linked glycosylation, and a signal peptide, with multiple isoforms produced by alternative splicing.
Experimental Guidance and Technical Tips
-
The immunogen (amino acids 19-256) encompasses the extracellular domain, consistent with detection of cell-surface ICOS ligand in flow cytometry (FC). Consider optimizing the antibody concentration by titration on a relevant cell line or primary cells known to express ICOSLG.
-
For FC, include appropriate PE-conjugated isotype controls to assess background, and use viable cell gating to exclude dead cells which may non-specifically bind the conjugate.
-
ICOSLG expression is inducible on activated monocytes and dendritic cells; thus, stimulation protocols may be required to detect the target in resting cell populations.
-
Predicted bands on western blot may appear at approximately 20, 33, and 34 kDa due to glycosylation and splice variants; however, this product is validated only for FC; verify suitability in other applications by independent testing.
