Biological Background: CD263/TRAIL-R3 Function and Localization
- Tumor necrosis factor receptor superfamily member 10C (TNFRSF10C), also known as Decoy Receptor 1 (DcR1), TRAIL-R3, CD263, LIT, and TRID.
- Functions as a receptor for the cytotoxic ligand TRAIL but lacks the intracellular death domain required to initiate apoptosis, thus acting as a decoy receptor.
- Protects cells from TRAIL-induced apoptosis by competing with the functional TRAIL receptors TNFRSF10A and TNFRSF10B for ligand binding.
- Localized to the cell membrane and anchored via a glycosylphosphatidylinositol (GPI) linkage.
- Higher expression in normal tissues compared to tumor cell lines; highly expressed in peripheral blood lymphocytes, spleen, skeletal muscle, placenta, lung, and heart.
- Contains disulfide bonds and is glycosylated; belongs to the TNF receptor superfamily characterized by cysteine-rich repeats.
Experimental Guidance and Technical Tips
- This antibody is supplied as a PE conjugate, allowing direct flow cytometry without the need for a secondary detection reagent.
- For optimal staining, titrate the antibody to determine the ideal concentration for your specific cell type and experimental conditions.
- As CD263/TRAIL-R3 is highly expressed in peripheral blood lymphocytes, these cells may serve as a suitable positive control for assay validation.
- Note that CD263/TRAIL-R3 is a GPI-anchored membrane protein; ensure that cell preparation and staining protocols preserve surface integrity.
- If using in combination with other antibodies, verify spectral compatibility of the PE fluorophore with your flow cytometer configuration.
