Biological Background: CD23 (Low affinity immunoglobulin epsilon Fc receptor) Function and Localization

  • Identified as the low affinity immunoglobulin epsilon Fc receptor (FcεRII), also known as CD23, BLAST-2, and C-type lectin domain family 4 member J (CLEC4J).
  • Functions as a receptor for IgE and for CR2/CD21, playing critical roles in regulating IgE production and B cell differentiation.
  • On B cells, CD23 mediates IgE-dependent antigen uptake and presentation to T cells, facilitating adaptive immune responses. Related references: PMID:2167225
  • On macrophages, upon IgE binding and antigen cross-linking, CD23 triggers intracellular killing of parasites via activation of the L-arginine-nitric oxide pathway. Related references: PMID:7544003
  • Encoded by the FCER2 gene, CD23 belongs to the C-type lectin superfamily, containing a carbohydrate recognition domain.
  • Subcellularly localized to the cell membrane as a type II transmembrane protein; also found as a secreted form (sCD23) generated by proteolytic cleavage.
  • Post-translational modifications include glycosylation, disulfide bond formation, and palmitoylation; the receptor binds calcium and IgE.
  • Tissue specificity: detected in urine at the protein level; broadly expressed on B cells, monocytes, and other immune cells.

Experimental Guidance and Technical Tips

  • The immunogen comprises the extracellular domain (aa 48–321) of human CD23, which includes the lectin and IgE-binding regions; this design likely targets functional epitopes relevant for receptor-ligand interaction studies.
  • For flow cytometry applications, validate the antibody’s performance in your specific experimental system, including appropriate isotype controls and Fc receptor blocking steps to minimize non-specific binding.
  • As a PE conjugate, ensure compensation settings are optimized when combining with other fluorophores; consider using a viability dye to exclude dead cells.
  • While validated for FC, other applications (e.g., IHC, Western blot) have not been tested and would require thorough optimization.
