Biological Background: CD221/IGF-1R Function and Localization

• Insulin-like growth factor 1 receptor (IGF1R) is a receptor tyrosine kinase that binds insulin-like growth factor 1 (IGF1) with high affinity, and IGF2 and insulin with lower affinity, mediating cell growth and survival control.
• Ligand binding triggers receptor autophosphorylation and phosphorylation of insulin receptor substrates (IRS1/2) and Shc, activating two major pathways: the PI3K-AKT/PKB pathway inhibits apoptosis and stimulates protein synthesis, while the Ras-MAPK pathway increases cellular proliferation.
• IGF1R also signals through the JAK/STAT pathway; phosphorylation of JAK proteins leads to activation of STAT proteins, particularly STAT3, which may be essential for the transforming activity of IGF1R.
• IGF1R can form hybrid receptors with the insulin receptor (INSR). Hybrid receptors composed of IGF1R and INSR isoform Long are activated with high affinity by IGF1, low affinity by IGF2, and not significantly activated by insulin. In contrast, another study shows that hybrid receptors with either INSR isoform Long or Short have similar binding characteristics, both binding IGF1 and having low affinity for insulin.
