Biological Background: IL13RA1 Function and Localization

  • IL13RA1 encodes the interleukin-13 receptor subunit alpha-1, also known as cancer/testis antigen 19 (CT19).
  • Binds IL13 with low affinity; together with IL4RA forms a functional receptor for IL13.
  • Serves as an alternate accessory protein to the common cytokine receptor gamma chain for IL4 signaling, but cannot replace IL2RG in allowing enhanced IL2 binding.
  • Ubiquitously expressed, with highest levels in heart, liver, skeletal muscle, and ovary; lowest in brain, lung, and kidney. Also present on B-cells, T-cells, and endothelial cells.
  • Membrane-localized receptor with a signal peptide, disulfide bonds, glycosylation, and a transmembrane helix.
  • Keywords: 3D-structure, Alternative splicing, Disulfide bond, Glycoprotein, Membrane, Proteomics identification, Receptor, Reference proteome, Repeat, Signal, Transmembrane, Transmembrane helix.

Experimental Guidance and Technical Tips

  • The immunogen encompasses the extracellular domain (amino acids 29-343), suggesting the antibody targets an extracellular epitope, suitable for flow cytometry with live unfixed cells.
  • Validate antibody reactivity with known CD213a1-expressing human cell lines or primary cells such as B-cells and T-cells.
  • Use appropriate isotype control antibodies conjugated to PE for accurate flow cytometry gating.
  • Since the antibody is PE-conjugated, ensure compensation controls are set up correctly for multicolor panels.
