Biological Background: Lymphocyte antigen 75 Function and Localization
- Encoded by the LY75 gene, lymphocyte antigen 75 (also known as DEC-205, CD205, CLEC13B, gp200-MR6) is a type I transmembrane protein belonging to the C-type lectin domain family. It exists in multiple isoforms due to alternative splicing, with isoform 2 and 3 being expressed in Hodgkin and Reed-Sternberg cells.
- Acts as an endocytic receptor that mediates the uptake of extracellular antigens and their delivery to a specialized antigen-processing compartment, thereby promoting antigen presentation and subsequent T cell activation. It also causes reduced proliferation of B-lymphocytes.
- Belongs to the C-type lectin domain family 13, characterized by a series of C-type lectin-like domains, which are involved in carbohydrate recognition and pathogen binding.
- Subcellular localization is primarily at the plasma membrane, consistent with its role as a cell-surface receptor.
- Post-translational modifications include glycosylation and phosphorylation. The protein contains disulfide bonds and a signal peptide, and is annotated as a glycoprotein and phosphoprotein.
- Exhibits tissue-specific expression in spleen, thymus, colon, and peripheral blood lymphocytes. Detected in myeloid and B-lymphoid cell lines, with isoform 2 and 3 specifically found in malignant Hodgkin lymphoma cells (Hodgkin and Reed-Sternberg cells).

- When working with human samples, validate the antibody in the relevant cell types known to express CD205, such as dendritic cells or B cells, and note that isoform-specific detection may differ in Hodgkin lymphoma.
