Biological Background: T-cell surface antigen CD2 Function and Localization

  • CD2 is a T-cell surface antigen also known as T11, LFA-2, SRBC (sheep red blood cell receptor), and the LFA-3 receptor. It belongs to the immunoglobulin superfamily.
  • It functions as a cell adhesion molecule by interacting with lymphocyte function-associated antigen CD58 (LFA-3) and CD48/BCM1, thereby mediating adhesion between T-cells and other cell types.
  • CD2 plays a role in T-cell activation and signaling; its cytoplasmic domain is implicated in signal transduction upon ligand binding.
  • The protein is a single-pass type I membrane glycoprotein localized at the cell membrane.
  • At the protein level, CD2 is expressed in natural killer cells and T lymphocytes.
  • Structural features include immunoglobulin-like domains, disulfide bonds, a signal peptide, a transmembrane helix, and extracellular glycosylation sites.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 25–209) of human CD2, which is expected to generate antibodies recognizing the native cell surface protein.
  • This antibody is directly conjugated to PE and optimized for flow cytometry; titrate the antibody to determine the optimal staining concentration for your sample.
  • Include appropriate isotype controls and compensation controls when performing multicolor flow cytometry.
  • Validate antibody performance in relevant positive cell lines or primary cells known to express CD2, such as Jurkat (T-cell) or NK cells.
  • Consider using a viability dye to exclude dead cells, as PE conjugates may exhibit non-specific binding.
