Biological Background: CD197/CCR7 Function and Localization

  • CD197/CCR7 is a C-C chemokine receptor type 7, also known as BLR2, CDw197, EBI1 (Epstein-Barr virus-induced G protein-coupled receptor 1), and MIP-3 beta receptor.
  • Functions as the receptor for the chemokine MIP-3-beta (CCL19), mediating lymphocyte chemotaxis and homing to secondary lymphoid organs.
  • Implicated in Epstein-Barr virus (EBV) effects on B-lymphocytes; expression is strongly upregulated in EBV-infected B-cells and herpesvirus-infected T-cells.
  • Belongs to the G protein-coupled receptor (GPCR) family, featuring seven transmembrane domains and a disulfide bond typical of chemokine receptors.
  • Localized at the cell membrane, consistent with its role as a transmembrane receptor and signal transducer.
  • Undergoes glycosylation, which may influence receptor stability, ligand binding, or trafficking.
  • Expressed in various lymphoid tissues and activated B- and T-lymphocytes, highlighting its role in adaptive immunity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal extracellular domain (aa 25-59), making this antibody suitable for surface staining in flow cytometry.
  • PE conjugation allows direct detection without secondary antibodies; titrate the antibody to establish optimal signal-to-noise ratio for your cell type.
  • As CCR7 is expressed on activated lymphocytes, include appropriate resting and activated lymphocyte controls.
  • For fixation/permeabilization, test compatibility as epitope may be sensitive.
  • Since the predicted molecular weight is 43 kDa, western blot may be possible but was not validated; if attempting, use reducing conditions and include positive controls.
