Biological Background: CCR6 Function and Localization
-
CCR6 (C-C chemokine receptor type 6) is a G protein-coupled receptor, also known as CD196, CKR-L3, DRY6, GPR29, STRL22, and CMKBR6, encoded by the CCR6 gene.
-
It functions as the primary receptor for the chemokine CCL20, and ligand binding induces intracellular calcium mobilization.

-
In addition to CCL20, CCR6 binds several beta-defensins (DEFB1, DEFB4, and DEFB4A/B), mediating chemotactic responses and regulating sperm motility and bactericidal activity.
Related references: PMID:20068036, PMID:25585877, PMID:25122636
-
The CCL20-CCR6 axis directs chemotaxis of dendritic cells, effector/memory T-cells, and B-cells, and is critical for immune surveillance at skin and mucosal surfaces, as well as in inflammatory diseases and cancer.
Related references: PMID:21376174
-
CCR6 is essential for the recruitment of both pro-inflammatory Th17 cells and regulatory T-cells (Treg) to sites of inflammation, thereby controlling T-cell balance in the intestine.
Related references: PMID:21376174
-
The receptor plays a role in early thymocyte development, B-cell homing to Peyer's patches, and IgA class switching; it also regulates sperm motility and chemotaxis via CCL20 binding.
Related references: PMID:23765988
-
CCR6 is localized at the cell surface and is expressed in lymphoid organs (spleen, lymph nodes, appendix), in lymphocyte subsets (T-cells, B-cells), and in the tail and postacrosomal region of sperm.
-
According to UniProt keywords, CCR6 is a transmembrane glycoprotein with a seven-helix structure, stabilized by disulfide bonds, and is implicated in G protein-mediated signal transduction.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular N-terminus (amino acids 1–40) of human CCR6, which may allow recognition of the native receptor on live cells in flow cytometry.
- For flow cytometric staining, use standard live-cell protocols with optimized PE-conjugated antibody concentrations and appropriate isotype controls.
- The PE fluorophore is compatible with common 561 nm or 488 nm laser lines; ensure instrument settings and compensation are adjusted for the experimental panel.
- Validate reactivity in relevant cell types, such as primary T helper cells, B cells, or CCR6-expressing cell lines, to confirm specificity and sensitivity.
