Biological Background: CD177 Function and Localization

  • CD177 (also known as NB1 glycoprotein, PRV-1, HNA-2a) is a GPI-anchored cell surface glycoprotein expressed on a subset of mature neutrophils.
  • In association with beta-2 integrin heterodimer ITGAM/CD11b and ITGB2/CD18, CD177 mediates activation of TNF-primed neutrophils, including degranulation and superoxide production. Related references: PMID:21193407
  • CD177 prevents beta-2 integrin internalization and attenuates chemokine signaling, switching the neutrophil response from migration to adhesion. Related references: PMID:28807980
  • Through heterophilic interaction with PECAM1 on endothelial cells, CD177 facilitates neutrophil transendothelial migration in vitro. Related references: PMID:17580308
  • Despite its role in transendothelial migration in vitro, CD177 appears to be dispensable for neutrophil recruitment during bacterial infection in vivo. Related references: PMID:23461681
  • CD177 acts as a receptor for the mature form of protease PRTN3, displaying it on the neutrophil cell surface. Related references: PMID:17244676 PMID:18462208
  • By presenting PRTN3 at the neutrophil surface, CD177 may enhance endothelial cell junctional integrity during neutrophil diapedesis. Related references: PMID:23202369
  • CD177 localizes to the cell membrane, membrane rafts, secretory vesicles, cytoplasmic granule membranes, and lamellipodia.
  • CD177 is highly expressed in normal bone marrow and in granulocytes from patients with polycythemia vera and essential thrombocythemia. Expression begins at the metamyelocyte stage during neutrophil differentiation, and the percentage of CD177-positive neutrophils varies widely among individuals.
  • The protein undergoes alternative splicing and possesses a GPI anchor, and its molecular weight varies among isoforms.

Experimental Guidance and Technical Tips

  • This antibody is generated against an immunogen spanning amino acids 22-407, corresponding to the extracellular domain of human CD177. This region is well-exposed on live cells, making the antibody suitable for flow cytometry detection of cell surface CD177.
  • For flow cytometry (FC), use appropriate isotype controls and titrate the antibody to determine optimal staining concentrations. Include a neutrophil gating strategy (e.g., CD16 and CD66b co-staining) to identify CD177-expressing populations.
  • Because CD177 is expressed on only a subset of neutrophils and expression levels vary between individuals, consider analyzing a cohort of donors and reporting percentage positivity rather than absolute cell numbers.
  • Pre-incubation with Fc receptor blocking reagents is recommended to minimize non-specific binding to Fcγ receptors on neutrophils.
  • This PE-conjugated antibody is ready-to-use; avoid repeated freeze-thaw cycles and protect from light to preserve fluorescence.
  • Cross-reactivity has been confirmed with human samples. For use with other species, validate binding and specificity independently.
