Biological Background: CD172a/SIRPα Function and Localization

  • Encoded by the SIRPA gene, this protein is a tyrosine-protein phosphatase non-receptor type substrate 1, also known as SIRPα, CD172a, and SHPS1. It belongs to the signal-regulatory protein family and exists as multiple isoforms (Sirp-alpha-1/2/3) due to alternative splicing.
  • Functions as an immunoglobulin-like cell surface receptor for CD47, acting as a docking protein that recruits and induces translocation of phosphatases PTPN6 and PTPN11 from the cytosol to the plasma membrane.
  • Negatively regulates phagocytosis, mast cell activation, and dendritic cell activation. CD47 binding prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells.
  • Supports adhesion of cerebellar neurons, promotes neurite outgrowth, and facilitates glial cell attachment, indicating a role in synaptogenesis and synaptic function (By similarity).
  • Involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors, or insulin.
  • Serves as a receptor for thrombospondin-1 (THBS1), leading to phosphorylation of SIRPα and subsequent ROS production through NADPH oxidase in non-phagocytic cells. Related references: PMID:24511121
  • Subcellular localization is at the membrane, consistent with its role as a transmembrane receptor. Post-translational modifications include phosphorylation and glycosylation.
  • Ubiquitously expressed, with highest levels in brain; also detected in heart, placenta, lung, liver, and various other tissues, but not on T-cells.
