Biological Background: CD170/Siglec-5 Function and Localization
- SIGLEC5 encodes Sialic acid-binding Ig-like lectin 5, also known as CD170, CD33 antigen-like 2 (CD33L2), and Obesity-binding protein 2 (OB-BP2).
- It functions as a putative adhesion molecule that mediates sialic-acid dependent binding to cells, recognizing both alpha-2,3- and alpha-2,6-linked sialic acid with equal affinity.
- The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface, potentially regulating its adhesive function.
- SIGLEC5 is a member of the sialic acid-binding immunoglobulin-like lectin (SIGLEC) family, characterized by an N-terminal V-set Ig domain and multiple C2-set Ig domains.
- Subcellularly, it is a transmembrane protein with a signal peptide, extracellular region, transmembrane helix, and cytoplasmic tail.
- Post-translational modifications include disulfide bonds and N-linked glycosylation, typical of cell surface receptors.
- Tissue specificity: Expressed by monocytic/myeloid lineage cells, with high levels in peripheral blood leukocytes, spleen, bone marrow, and lower levels in lymph node, lung, appendix, placenta, pancreas, and thymus. It is present on monocytes and neutrophils but absent from early-stage leukemic cell lines.
- The mature protein has a predicted molecular weight of approximately 61 kDa.
Experimental Guidance and Technical Tips
- This antibody is directly conjugated to PE, allowing immediate use in flow cytometry without a secondary detection step. Ensure appropriate compensation controls when using multiple fluorophores.
- The immunogen corresponds to the extracellular region (amino acids 17-434), targeting the sialic acid-binding domain. This region is expected to be accessible on live, non-permeabilized cells.
- For flow cytometry, validate staining on human peripheral blood monocytes or neutrophils, which naturally express CD170/Siglec-5. Consider using a myeloid cell line as a positive control.
- Because cis-interactions with cell-surface sialic acids can mask the epitope, consider treating cells with sialidase prior to staining to unmask CD170/Siglec-5, especially if weak signals are observed.
- Use an appropriate PE-conjugated rabbit isotype control to distinguish specific binding from background.
- The antibody is validated for human reactivity; cross-reactivity with other species has not been confirmed. Test on samples from the desired species if used beyond human.
