Biological Background: CD161 (KLRB1) Function and Localization
- KLRB1 encodes Killer cell lectin-like receptor subfamily B member 1, also known as CD161, NKR-P1A, and CLEC5B.
- CD161 plays an inhibitory role on natural killer (NK) cell cytotoxicity. Activation stimulates acid sphingomyelinase/SMPD1, elevates intracellular ceramide, and enhances T-cell proliferation.
- It acts as a lectin that binds to the terminal Gal-alpha(1,3)Gal epitope and N-acetyllactosamine. CD161 also binds to its ligand CLEC2D/LLT1, inhibiting NK cell-mediated cytotoxicity and interferon-gamma secretion in target cells.
- CD161 is a membrane protein with a signal-anchor and transmembrane helix, classified under the C-type lectin domain family.
- Expressed in a subset of NK cells, predominantly in intestinal epithelium and liver, and detected in peripheral blood T-cells, preferably with a memory antigenic phenotype.
- Post-translational modifications include glycosylation and disulfide bond formation.
- The protein has a calculated molecular weight of approximately 25 kDa.
