Biological Background: ADAM10 Function and Localization
- ADAM10 (Disintegrin and metalloproteinase domain-containing protein 10), also known as CD156c and Kuzbanian, is a transmembrane metalloprotease belonging to the ADAM family.
- It functions as a major ectodomain sheddase for a wide range of adhesion proteins, growth factor precursors, and cytokines, playing essential roles in development and tissue homeostasis. Related references: PMID:11786905, PMID:12475894, PMID:20592283
- ADAM10 cleaves the membrane-bound precursor of tumor necrosis factor (TNF) and is responsible for the constitutive and regulated alpha-secretase cleavage of amyloid precursor protein (APP), linking it to inflammation and Alzheimer's disease pathology. Related references: PMID:11786905, PMID:26686862, PMID:29224781
- It mediates the proteolytic release of cellular prion protein and adhesion molecule L1, and controls Notch receptor processing, which is critical for neurogenesis and immune cell development. Related references: PMID:11477090, PMID:12475894
- ADAM10 sheds FasL, generating a transmembrane signaling remnant, and cleaves the immune checkpoint receptor LAG3, influencing immune regulation. Related references: PMID:17557115
- The protein localizes to the cell membrane, Golgi apparatus, cytoplasmic vesicles, and adherens junctions, with expression detected in the brain, spleen, lymph nodes, and peripheral blood leukocytes.
- As a zinc-dependent metalloprotease, ADAM10 is characterized by disulfide bonds, glycosylation, and phosphorylation, and its dysregulation is associated with Alzheimer disease and amyloidosis.
Experimental Guidance and Technical Tips
- This PE-conjugated antibody is suitable for direct flow cytometry to detect surface ADAM10 on live human cells.
- We recommend titrating the antibody to determine the optimal concentration for your specific cell type and assay conditions.
- Include appropriate isotype control and Fc receptor blocking to minimize non-specific binding.
- If intracellular staining is required, validate fixation and permeabilization protocols for compatibility with PE fluorescence.
- Confirm ADAM10 expression in your target cell population using known positive and negative controls.
