Biological Background: CD131/CSF2RB Function and Localization
- The CSF2RB gene encodes the cytokine receptor common subunit beta, also known as CD131, CDw131, IL3RB, IL5RB, and GM-CSF/IL-3/IL-5 receptor common beta subunit.
- It forms heterodimers with specific alpha subunits (IL3RA, IL5RA, or CSF2RA) to mediate signaling by interleukin-3, interleukin-5, and granulocyte-macrophage colony-stimulating factor (CSF2), regulating hematopoietic progenitor cell differentiation. Related references: PMID:1495999
- Under basal conditions, CSF2RB constitutively associates with JAK1. Ligand binding triggers JAK1 activation and subsequent phosphorylation cascade through the JAK-STAT pathway. Related references: PMID:9516124
- The receptor is a single-pass type I membrane glycoprotein, localized to the plasma membrane (SUBCELLULAR LOCATION: Membrane; KEYWORDS: Transmembrane, Transmembrane helix).
- CSF2RB undergoes post-translational modifications including glycosylation and phosphorylation, and contains disulfide bonds critical for its structure. Alternative splicing generates multiple isoforms.
- It plays a central role in immune responses by controlling the production and activity of eosinophils, basophils, and other myeloid cells.
- The canonical isoform has a predicted molecular weight of approximately 97.3 kDa, consistent with the observed band at 97 kDa in product validation.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 17-443) covers the extracellular domain; consider epitope mapping if precise binding site is required.
- For flow cytometry, titrate the antibody to determine the optimal staining concentration for your cell type and experimental conditions.
- Use appropriate compensation controls and avoid prolonged light exposure when handling PE-conjugated antibodies.
- Validate assay performance using human peripheral blood mononuclear cells (PBMCs) or relevant hematopoietic cell lines known to express CD131.
- Cross-reactivity has been confirmed for human; if testing other species, check sequence homology of the target protein.
