Biological Background: CD124/IL-4Rα Function and Localization

  • CD124 (IL-4Rα) is a receptor subunit for both interleukin 4 (IL-4) and interleukin 13 (IL-13), playing a central role in type 2 immune responses. Related references: PMID:17030238
  • Upon ligand binding, it couples to the JAK1/2/3-STAT6 signaling cascade, driving transcriptional programs associated with Th2 cell differentiation.
  • In B cells and other cell types, IL-4/IL-13 signaling through IL-4Rα regulates IgE class switching and production, as well as chemokine and mucus secretion at sites of allergic inflammation.
  • In certain cell contexts, IL-4Rα can also signal via insulin receptor substrates IRS1 and IRS2, linking cytokine signaling to metabolic pathways.
  • A soluble isoform (sIL4R) is produced by alternative splicing and acts as a decoy receptor, inhibiting IL-4-mediated proliferation and IL-5 upregulation in T cells.
  • The receptor is primarily localized to the cell membrane, with the soluble form being secreted; it is highly expressed in activated T-cells (isoforms 1 and 2).
