Biological Background: CD117/c-Kit Function and Localization
- CD117, also known as c-Kit, SCFR, or PBT, is a transmembrane tyrosine-protein kinase receptor encoded by the KIT gene; alternative splicing produces multiple isoforms (UniProt: P10721).
- The primary ligand is stem cell factor (SCF/KITLG). Ligand binding induces receptor dimerization and autophosphorylation, activating downstream signaling cascades that regulate cell survival, proliferation, and differentiation in hematopoiesis, melanogenesis, gametogenesis, and mast cell development.
- Activated CD117 recruits and phosphorylates key adaptor proteins and enzymes including PIK3R1 (PI3K), GRB2, and PLCG1, leading to activation of AKT, RAS/MAPK, and STAT (STAT1, STAT3, STAT5) pathways, as well as production of second messengers DAG and IP3.
- CD117 is predominantly localized on the cell membrane, but can also be found in the cytoplasm upon internalization and trafficking.
- In testis, CD117 is detected in spermatogonia in the basal layer and in interstitial Leydig cells, and its expression is maintained in ejaculated spermatozoa (at protein level). Related references: PMID:20601678
- The protein is heavily glycosylated, contains extracellular immunoglobulin domains and an intracellular kinase domain, and is classified as a proto-oncogene; disulfide bonds and phosphorylation are key structural and functional features.
