Biological Background: Calreticulin Function and Localization
- Calreticulin (CALR, also known as CRP55, Calregulin, ERp60, HACBP, grp60) is a highly conserved calcium-binding chaperone primarily localized to the endoplasmic reticulum (ER) lumen.
- It serves as a central component of the calreticulin/calnexin cycle, promoting the folding, oligomeric assembly, and quality control of newly synthesized glycoproteins by binding to monoglucosylated N-linked glycans (PubMed:7876246). Related references: PMID:7876246
- Calreticulin interacts with the DNA-binding domain of the glucocorticoid receptor (NR3C1) and mediates its nuclear export, thereby linking ER function to cytoplasmic signaling pathways (PubMed:11149926). Related references: PMID:11149926
- Beyond the ER, calreticulin localizes to the cytosol, cell surface, extracellular matrix, and secretory vesicles; it is also detected in the sarcoplasmic reticulum, cortical granules, and cytolytic granules, reflecting its multifunctional roles.
- It participates in calcium homeostasis, cellular adhesion, and gene expression regulation, and has been implicated in oocyte maturation and the block to polyspermy.
- Post-translational modifications include acetylation, hydroxylation, and a conserved disulfide bond; the protein has a molecular weight of approximately 48 kDa and contains a signal peptide, lectin-binding domains, and metal-binding sites.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (amino acids 335–417) of human calreticulin; verify that this epitope is accessible in your specific fixation and permeabilization protocol for intracellular flow cytometry.
- Since this antibody is PE-conjugated and recommended for intracellular staining, consider optimizing permeabilization conditions and including appropriate isotype controls to account for potential background due to intracellular Fc receptors or non-specific binding.
- Calreticulin is expressed in multiple subcellular compartments; validate staining specificity by comparing localization patterns in the ER, cytosol, or cell surface, depending on your experimental context.
- For applications other than intracellular flow cytometry, pilot testing is strongly recommended, as this antibody is validated only for the stated application under the supplier’s conditions.
