Biological Background: C5AR1 Function and Localization
- C5AR1 (C5a anaphylatoxin chemotactic receptor 1, also known as C5a-R, C5aR, CD88) is a G protein-coupled receptor that serves as the receptor for the chemotactic and inflammatory peptide anaphylatoxin C5a.
- Upon C5a binding, C5AR1 undergoes a conformational change that triggers signaling via guanine nucleotide-binding proteins (G proteins), primarily coupled to G(i)/G(o) alpha subunits (GNAI1 or GNAO1), leading to inhibition of adenylate cyclase. Related references: PMID:36806352, PMID:37852260
- Activation of C5AR1 stimulates a range of cellular responses including chemotaxis, granule enzyme release, intracellular calcium release, and superoxide anion production, implicating it in inflammatory processes. Related references: PMID:10636859, PMID:15153520, PMID:1847994
- The ligand C5a interacts with at least two sites on C5AR1: a high-affinity site on the extracellular N-terminus and a second site within the transmembrane region that activates downstream signaling. Related references: PMID:7622471, PMID:8182049, PMID:9553099
- Subcellularly, C5AR1 localizes to the cell membrane and cytoplasmic vesicles, consistent with its roles in signal transduction and receptor trafficking.
- C5AR1 is post-translationally modified by glycosylation, phosphorylation, and sulfation, and contains disulfide bonds characteristic of GPCRs, as indicated by UniProt keywords.
- As a member of the G protein-coupled receptor family, C5AR1 functions as a transducer in chemotactic signaling pathways and is identified in proteomic studies.
Experimental Guidance and Technical Tips
- This antibody was generated against a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human C5AR1. For optimal performance in flow cytometry (FC), consider titrating the antibody to determine the appropriate working concentration for your specific cell type and experimental conditions.
- Due to the receptor's expression on cell membranes and intracellular vesicles, permeabilization steps may affect staining patterns; evaluate both surface and total staining protocols as needed.
- Validate the antibody specificity using a relevant positive control cell line known to express C5AR1 (e.g., immune cells) and a negative control (e.g., receptor-negative cells or isotype control).
- If using this PE-conjugated antibody in multicolor flow panels, ensure proper compensation and avoid potential spectral overlap with other fluorophores.
