Biological Background: CD15/SSEA-1 Function and Localization

  • FUT4 (fucosyltransferase 4) is a member of the glycosyltransferase family, responsible for transferring fucose to N-acetylglucosamine on glycoproteins, thereby generating the Lewis X (CD15) carbohydrate antigen.
  • Catalyzes the alpha(1→3) linkage of fucose to distal N-acetylglucosamine of type 2 lactosamine chains, robustly forming Lewis X (CD15), and with lower efficiency, sialyl Lewis X and 6-sulfo sialyl Lewis X—key selectin recognition determinants (PubMed:1702034, PubMed:1716630, PubMed:29593094).   • Together with FUT7, contributes to the biosynthesis of selectin ligands (e.g., SELE, SELL, SELP), mediating lymphocyte homing, leukocyte extravasation, and immune cell homeostasis (By similarity).
