Biological Background: CD11b Function and Localization
- Integrin alpha-M (ITGAM/CD11b) is a leukocyte integrin alpha chain that pairs with integrin beta-2 (ITGB2/CD18) to form the Mac-1 (CR-3) complex.
- ITGAM/ITGB2 mediates adhesive interactions of monocytes, macrophages, and granulocytes and serves as the CR-3 receptor for the iC3b fragment of complement component C3, promoting uptake of complement-coated particles and pathogens. Related references: PMID:20008295, PMID:9558116
- ITGAM/ITGB2 acts as a receptor for fibrinogen, factor X, and ICAM1; ICAM1 binding facilitates adhesion between stimulated neutrophils and endothelial cells. Related references: PMID:1980124
- The complex is required for CD177-PRTN3-mediated activation of TNF-primed neutrophils and regulates neutrophil migration. Related references: PMID:21193407, PMID:28807980
- Predominantly expressed in monocytes and granulocytes, with protein expression confirmed in neutrophils; localizes to the cell membrane and membrane rafts. Related references: PMID:1346576, PMID:21193407
- Structural features include a signal peptide, multiple extracellular integrin alpha repeats, a transmembrane helix, disulfide bonds, and N-glycosylation sites.
- Beyond adhesion, ITGAM/ITGB2 may contribute to phagocytosis-induced apoptosis in extravasated neutrophils, mast cell development, and, in microglia, TYROBP/DAP12-dependent superoxide production that promotes neuronal apoptosis during brain development.
- Genetic variants in ITGAM are associated with systemic lupus erythematosus (SLE), underscoring its role in immune regulation.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions.
- As the immunogen is a synthetic peptide, the epitope is likely linear; the antibody may be compatible with both live cells and fixed/permeabilized protocols, but validation in your system is recommended.
- Use appropriate compensation controls (e.g., single-stained cells or compensation beads) to correct for spectral overlap of PE with other fluorophores.
- Although specificity is reported for human CD11b, cross-reactivity with other species should be verified if needed; consider sequence alignment of the immunogen region.
