Biological Background: CD22 Function and Localization
- CD22 (Siglec-2, BL-CAM) is a sialic acid-binding immunoglobulin-like lectin predominantly expressed on B-cells, with alternative splicing yielding multiple isoforms (predicted molecular weights 89–97 kDa).
- It plays essential roles in B-cell biology, including differentiation, antigen presentation, and bone marrow trafficking.

PMID:7533044
PMID:9016707 - CD22 binds α2,6-linked sialic acids on cis ligands (e.g., CD22, CD45, IgM) and trans ligands on opposing cells, mediating cell adhesion.
- As an inhibitory coreceptor, it dampens BCR signaling: upon LYN-mediated ITIM phosphorylation, it recruits SHP-1 phosphatase, inhibiting calcium mobilization and cell activation. Related references: PMID:9016707
- Subcellularly, CD22 is localized to the plasma membrane of B-lymphocytes.
- The protein features immunoglobulin domains, disulfide bonds, and regulatory phosphorylation sites.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 22-151) of mouse CD22, which may recognize both cis- and trans-interacting forms; validate specificity in appropriate model systems.
- For flow cytometry, titrate the antibody to determine optimal staining concentration; include appropriate isotype controls and consider Fc receptor blocking.
- As a PE-conjugated antibody, ensure proper compensation when using multicolor panels; protect from light to prevent fluorophore bleaching.
