Biological Background: BCL6 Function and Localization
- BCL6 is a transcriptional repressor primarily required for germinal center formation and antibody affinity maturation.
- It binds directly to the DNA sequence 5'-TTCCTAGAA-3' (BCL6-binding site) and forms complexes with corepressors and histone deacetylases to repress target genes.
- In germinal center B-cells, BCL6 represses genes involved in differentiation, inflammation, apoptosis, and cell cycle control; it also indirectly upregulates AICDA through microRNA repression.
- BCL6 promotes follicular helper T-cell gene expression while inhibiting differentiation of Th1, Th2, and Th17 cells, and is essential for immunological memory in both T- and B-cells.
- The protein localizes to the nucleus and is highly expressed in germinal center T- and B-cells, as well as in primary immature dendritic cells.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitination; it features zinc-finger domains and is classified as a proto-oncogene.
- Additionally, BCL6 regulates neurogenesis by altering NOTCH-dependent transcriptional complexes and recruiting SIRT1 to silence selective NOTCH targets.
