Biological Background: CD38 Function and Localization

  • CD38 (ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1) is a multifunctional transmembrane glycoprotein also known as I-19 antigen and NIM-R5 antigen. It belongs to the ADP-ribosyl cyclase family.
  • CD38 synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion, and possesses cADPR hydrolase activity.
  • CD38 catalyzes the synthesis of the Ca²⁺ mobilizer nicotinate-adenine dinucleotide phosphate (NAADP⁺) from 2'-phospho-cADPR and nicotinic acid, as well as from NADP⁺ and nicotinic acid (PubMed:11829748). Related references: PMID:11829748
  • CD38 functions as a receptor that binds CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and migration across the endothelial barrier.
  • CD38 is located on the membrane, with a signal-anchor transmembrane domain.
  • The protein contains disulfide bonds, is glycosylated, and has a molecular weight of approximately 34 kDa.
  • CD38 is involved in regulating dendritic cell functions, including chemotaxis, survival, and T-cell polarization.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 45–304 of mouse CD38, covering the extracellular domain; this design may favor recognition of the native conformation.
  • For flow cytometry applications, titrate the PE-conjugated antibody to determine the optimal staining concentration for your specific cell type and assay conditions.
  • Include appropriate isotype controls and viability dyes to ensure accurate gating.
  • Validate the antibody in a relevant mouse cell system, such as splenocytes or CD38-expressing cell lines, before large-scale experiments.
