Biological Background: CD27 Function and Localization
- CD27 (also known as TNFRSF7) is a costimulatory immune-checkpoint receptor of the tumor necrosis factor receptor superfamily, expressed on T-cells, NK-cells, and B-cells.
- Binding to its ligand CD70/CD27L triggers CD27-mediated T-cell activation and expansion, which in turn facilitates immune surveillance of B-cells. Related references: PMID:28011863
- Mechanistically, CD70 ligation activates the TRAF2-PTPN6 signaling axis, leading to inhibition of LCK phosphorylation and promoting T-cell memory differentiation. Related references: PMID:38354704
- CD27 also negatively regulates Th2 lymphocyte function in adipose tissue and provides a negative feedback signal to leukocyte differentiation during immune activation.
