Biological Background: CD209/DC-SIGN Function and Localization

  • CD209, also known as DC-SIGN (Dendritic Cell-Specific ICAM-3-Grabbing Non-integrin), is a C-type lectin receptor encoded by the CD209 gene (alias CLEC4L). It is a pathogen-recognition receptor expressed on immature dendritic cells.
  • CD209 functions as a high-affinity receptor for ICAM2 and ICAM3 via calcium-dependent mannose-like carbohydrate binding, mediating dendritic cell transendothelial migration and T-cell clustering. Related references: PMID:10721994, PMID:10721995, PMID:11017109
  • It is involved in the recognition and internalization of a broad spectrum of pathogens, including viruses (HIV-1, HIV-2, Ebola, HCV, Dengue, Measles, HSV-1, Influenza A, SARS-CoV, Japanese encephalitis, Lassa, Marburg, RSV, Rift Valley fever, Uukuniemi, West Nile) and bacteria (M. tuberculosis, H. pylori, K. pneumoniae). Related references: PMID:23966408, PMID:16092920, PMID:21203928, PMID:16379498
  • CD209 mediates endocytosis of pathogens and presents antigens to T cells via MHC class II, initiating adaptive immune responses.
  • Predominantly expressed in dendritic cells and DC-residing tissues, with additional expression in placental macrophages, endothelial cells, PBMCs, and THP-1 monocytes.
  • The protein exists in multiple isoforms (due to alternative splicing) and is localized to membrane rafts, cell membrane, and secreted forms.
  • It is a glycoprotein with calcium-dependent mannose-binding activity and contains a C-type lectin domain, a signal-anchor, and transmembrane region.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular region (aa 59-404) encompassing the carbohydrate-recognition domain (CRD) of CD209; this region is critical for ligand binding and pathogen interaction, making it suitable for detecting functional CD209 in flow cytometry.
  • For flow cytometry, titrate the antibody using relevant cell lines (e.g., dendritic cells, THP-1 monocytes) to determine optimal staining concentration. Include appropriate isotype controls.
  • Consider fixation and permeabilization steps only if intracellular staining is intended; CD209 is primarily cell surface-expressed but may also exist in intracellular compartments.
  • Validate the antibody in your experimental system, as post-translational modifications (e.g., glycosylation) might affect epitope accessibility.
