Biological Background: FUT4 (CD15/SSEA-1) Function and Localization
- FUT4, also known as fucosyltransferase IV (FucT-IV), is an alpha-(1,3)-fucosyltransferase that catalyzes the transfer of fucose to N-acetylglucosamine (GlcNAc) in type 2 lactosamine chains.
- Robustly fucosylates nonsialylated distal LacNAc units to form the Lewis X antigen (CD15), a key glycan determinant in development and immunity. Related references: PMID:1702034, PMID:1716630, PMID:29593094
- With lower efficiency, modifies sialylated LacNAc to generate sialyl Lewis X and 6-sulfo sialyl Lewis X determinants, which serve as selectin recognition epitopes. Related references: PMID:1716630, PMID:29593094
- Together with FUT7, contributes to selectin ligand biosynthesis and leukocyte trafficking.
- In certain cell types, may also fucosylate internal LacNAc units to create the VIM-2 antigen, another SELE epitope. Related references: PMID:11278338, PMID:1716630
- Does not generate Lewis X antigens.
- Localizes to the Golgi stack membrane.
- Expressed in bone marrow mesenchymal stem cells, cord blood promyelocytes, and peripheral blood myeloid and lymphoid cells.
