- PDK4, or [pyruvate dehydrogenase (acetyl-transferring)] kinase isozyme 4, is a mitochondrial kinase that phosphorylates and inactivates pyruvate dehydrogenase subunits PDHA1 and PDHA2, thereby regulating the tricarboxylic acid cycle and down-regulating aerobic respiration.
- By inhibiting pyruvate dehydrogenase, PDK4 shifts cellular metabolism from glucose utilization to fatty acid oxidation, playing a crucial role in metabolic homeostasis during prolonged fasting and starvation.
- PDK4 is involved in maintaining normal blood glucose levels, participating in the insulin signaling cascade, and preventing excessive ketone body accumulation under starved conditions.
- In the fed state, PDK4 mediates cellular responses to glucose levels and high-fat diet, balancing fatty acid oxidation and de novo fatty acid biosynthesis.
