Biological Background: PCGF3 Function and Localization

• PCGF3, also known as RING finger protein 3A (RNF3A), is a member of the polycomb group (PcG) RING finger protein family. It is encoded by the PCGF3 gene, which undergoes alternative splicing to produce multiple isoforms.
• PCGF3 functions as a component of the PRC1-like complex, which is essential for maintaining the transcriptionally repressive state of many genes, including Hox genes, throughout development. This complex acts via chromatin remodeling and mediates the monoubiquitination of histone H2A at lysine 119 (H2AK119ub).

• PCGF3 plays a redundant role with PCGF5 in mediating monoubiquitination of H2AK119 on the X chromosome, a process required for normal X-chromosome inactivation in XX female cells.

• For Western blotting, a band migrating at approximately 28 kDa is expected for PCGF3. However, due to potential post-translational modifications or isoform variants, additional bands may appear. Validate the antibody in the relevant sample system, preferably using a positive control cell line or tissue expressing PCGF3.
• In ELISA, the polyclonal nature of this antibody may allow it to serve as both capture and detection reagent, but pairing with a monoclonal antibody is recommended for sandwich ELISA. Titrate antibody concentrations and validate with recombinant protein or endogenous samples.
• Cross-reactivity with mouse is reported, enabling the use of this antibody in mouse models. When working with mouse samples, consider potential background and include appropriate species-matched controls.
