Biological Background: PARP3 Function and Localization
- PARP3 (also known as ARTD3, IRT1) is a member of the poly(ADP-ribose) polymerase family and functions as a mono-ADP-ribosyltransferase, unlike PARP1 and PARP2 which catalyze poly-ADP-ribosylation (PubMed:25043379). Related references: PMID:25043379
- It plays a critical role in the DNA damage response by mediating mono-ADP-ribosylation of target proteins at glutamate, aspartate, or lysine residues, facilitating repair of DNA strand breaks (PubMed:16924674, PubMed:20064938). Related references: PMID:16924674, PMID:20064938
- PARP3 cooperates with the Ku70/Ku80 heterodimer (XRCC5/XRCC6) to limit DNA end-resection and promote accurate non-homologous end joining (NHEJ), and it ADP-ribosylates histones such as H2B on nucleosomes at damage sites (PubMed:24598253, PubMed:27530147). Related references: PMID:24598253, PMID:27530147
- It also catalyzes DNA mono-ADP-ribosylation at strand break termini, adding a single ADP-ribose moiety to 5’- or 3’-phosphate groups (PubMed:29361132, PubMed:29520010). Related references: PMID:29361132, PMID:29520010
- Additionally, PARP3 associates with repair factors (e.g., APLF) to retain the LIG4-XRCC4 complex on chromatin, accelerating ligation during NHEJ (PubMed:21211721). Related references: PMID:21211721
- Predominantly nuclear with chromosome association; also detected at centrosomes and centrioles, potentially linking DNA damage to mitotic checkpoints (PubMed:16924674). Related references: PMID:16924674
- PARP3 is widely expressed in human tissues, with highest levels in kidney, skeletal muscle, liver, heart, and spleen.
Experimental Guidance and Technical Tips
- The recombinant immunogen corresponding to amino acids 178-533 of human PARP3 (NP_005476.4) suggests suitability for detecting the full-length protein (~60 kDa) by Western blot; consider validating in human, mouse, or rat cell lysates as cross-reactivity is indicated.
- For IHC-P and IF/ICC, optimize fixation and antigen retrieval conditions, as nuclear and centrosomal localization may require specific permeabilization steps.
- In ELISA applications, the antibody may serve as a capture or detection reagent; performance should be titrated against target antigen.
