Biological Background: PARP10 Function and Localization

  • PARP10, also known as ARTD10, is a member of the poly(ADP-ribose) polymerase family that functions as a mono-ADP-ribosyltransferase.
  • It catalyzes the transfer of a single ADP-ribose unit to glutamate and aspartate residues, and unlike PARP1 and PARP2, lacks poly-ADP-ribosylation activity. Related references: PMID:18851833, PMID:23332125, PMID:23474714
  • PARP10 mono-ADP-ribosylates GSK3B, thereby negatively regulating GSK3B kinase activity. Related references: PMID:23332125
  • In response to DNA damage, PARP10 interacts with PCNA to facilitate translesion DNA synthesis. Related references: PMID:24695737
  • The protein localizes to the nucleus, nucleolus, and cytoplasm.
  • Tissue expression is highest in spleen and thymus, with intermediate levels in liver, kidney, pancreas, prostate, testis, ovary, intestine, and leukocytes, and low expression in heart, brain, placenta, lung, skeletal muscle, and colon.
  • Post-translational modifications include acetylation, phosphorylation, and auto-ADP-ribosylation, which modulate its function.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-300) of human PARP10. In WB, a band near 110 kDa is expected for the full-length protein; confirm specificity using knockout controls or siRNA knockdown.
  • Spleen and thymus lysates are recommended as positive controls due to high PARP10 expression; also consider relevant cell lines.
  • For ELISA, coating with recombinant PARP10 protein or immunogen fragment and standard indirect protocol should be optimized.
  • Titrate primary antibody concentration (e.g., 1:500 to 1:2000 for WB) and incubation conditions for optimal signal.
  • When working with non-human species, verify cross-reactivity experimentally as this antibody is validated for human.
