Biological Background: PRKC apoptosis WT1 regulator protein Function and Localization
- PRKC apoptosis WT1 regulator protein (also known as Prostate apoptosis response 4 protein, Par-4) is encoded by the PAWR gene and functions as a pro-apoptotic factor that selectively induces apoptosis in cancer cells.
- The protein activates the Fas prodeath pathway and simultaneously inhibits NF-kappa-B transcriptional activity, promoting tumor regression in animal models.
- It interacts with WT1 to inhibit transcriptional activation and augment repression, leading to down-regulation of the anti-apoptotic protein BCL2.
- PAR4 may also act as a direct transcriptional repressor and is implicated in regulating amyloid precursor protein (APP) cleavage by BACE1.
- Subcellularly, the protein is localized to both the cytoplasm and nucleus, consistent with its dual roles in signaling and transcription regulation.
- Widely expressed in tissues, its levels are elevated in neurodegenerative diseases (ALS, Alzheimer's, Parkinson's, Huntington's) and stroke, while down-regulated in several cancers.
- Keywords from UniProt: 3D-structure, Apoptosis, Coiled coil, Cytoplasm, Nucleus, Phosphoprotein, Proteomics identification, Reference proteome, Transcription, Transcription regulation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from amino acids 241-340 of human PAR4, which lies in the central region; this may offer distinct epitope recognition compared to N- or C-terminal antibodies.
- For Western blot (WB), expect a band at approximately 37 kDa; use appropriate reducing conditions and validate in cancer cell lines where PAR4 is downregulated.
- Immunohistochemistry (IHC-P) may require antigen retrieval and should be validated with tissue controls comparing normal and tumor samples due to differential expression.
- ELISA applications can be developed for quantitative detection; cross-reactivity with both human and mouse PAR4 allows potential use in preclinical mouse models.
- Consider pairing with other apoptosis markers (e.g., BCL2, Fas) in multi-analyte assays to study PAR4-mediated pathways.
