Biological Background: PAI-1/Serpin E1 Function and Localization
- PAI-1, encoded by the SERPINE1 gene, is a secreted serine protease inhibitor (serpin) that also functions as a regulator of cell migration independently of its protease inhibitory activity. Related references: PMID:15001579, PMID:9168821
- It is a principal inhibitor of tissue-type plasminogen activator (PLAT), essential for down-regulating fibrinolysis and controlling blood clot degradation. Related references: PMID:17912461, PMID:8481516, PMID:9207454
- PAI-1 inhibits urokinase-type plasminogen activator (PLAU), thereby modulating cell adhesion and spreading. Related references: PMID:9175705
- It is required for stimulation of keratinocyte migration during cutaneous injury repair. Related references: PMID:18386027
- PAI-1 is involved in cellular and replicative senescence, including alveolar type 2 cell senescence. Related references: PMID:16862142
- It plays a role in cementogenic differentiation of periodontal ligament stem cells and in odontoblast differentiation during dentin formation. Related references: PMID:25808697, PMID:27046084
- Subcellularly, PAI-1 is secreted and found in plasma, platelets, and endothelial cells, with expression also observed in hepatoma and fibrosarcoma cells. Related references: PMID:2430793, PMID:3097076
- PAI-1 is a glycoprotein with a predicted molecular weight of approximately 45 kDa and undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 50-150 of human PAI-1, indicating the antibody may recognize both native and denatured protein. For WB, consider using reducing conditions and validate in human or monkey cell lysates as cross-reactivity data suggest.
- For ELISA, optimization of antibody dilution and incubation conditions is recommended based on the specific assay format and sample type. Secreted PAI-1 can be detected in cell supernatants or biological fluids.
- Since the antibody is monoclonal, lot-to-lot consistency is expected; however, it is advisable to titrate the antibody for each new application or sample system.
