Biological Background: OsSERK2 Function and Localization
- Also known as OsSERK1 or OsBAK1-2, OsSERK2 is a member of the somatic embryogenesis receptor kinase (SERK) family in rice.
- Involved in positive regulation of somatic embryogenesis and defense response against the rice blast fungus Magnaporthe oryzae. Related references: PMID:15968510
- Required for immunity mediated by the LRR receptor kinases XA21 and XA26/XA3 against Xanthomonas oryzae pv. oryzae, with kinase activity and direct interaction critical for function. Related references: PMID:24482436
- Necessary for the immune response mediated by the LRR receptor kinase FLS2 upon recognition of bacterial flagellin (flg22). Related references: PMID:24482436
- Participates in brassinosteroid (BR) signaling pathway regulating plant growth. Related references: PMID:19754838 PMID:24482436
- Expressed in flag leaves, roots, shoot apex, leaf blades, leaf sheaths, panicles, and flowers. Related references: PMID:11332734 PMID:16081169 PMID:24482436
- Localized to the cell membrane, consistent with its receptor kinase function.
- Post-translationally modified by glycosylation and phosphorylation, as indicated by UniProt keywords.
- Belongs to the SERK family of plant receptor kinases, characterized by an extracellular leucine-rich repeat (LRR) domain, a transmembrane helix, and an intracellular serine/threonine kinase domain.
Experimental Guidance and Technical Tips
- The immunogen used to raise this antibody is a recombinant protein of Oryza sativa OsSERK2. Verify specificity by using positive and negative control tissue lysates in WB.
- For Western blot, a band around 70 kDa is expected; however, post-translational modifications (glycosylation, phosphorylation) may cause slight shifts. Optimize blocking and detection conditions for rice samples.
- This antibody has been validated for ELISA; consider using it as a capture or detection antibody in sandwich assays after pairing tests.
- Because cross-reactivity is reported only for Oryza sativa, validate reactivity in other plant species if broader use is intended.
