Biological Background: Oncostatin M Function and Localization
- Oncostatin M (OSM) is a secreted glycoprotein belonging to the interleukin-6 (IL-6) family of cytokines, characterized by conserved disulfide bonds and cleavage on pair of basic residues.
- It exerts its biological activities through binding to two distinct receptor complexes: the type I receptor (heterodimer of LIFR and IL6ST/gp130) and the type II receptor (heterodimer of OSMR and IL6ST/gp130). Related references: PMID:10997905 PMID:39532904 PMID:8999038
- Upon ligand binding, receptor dimerization activates associated JAK kinases (JAK1, JAK2, TYK2), which phosphorylate receptor intracellular domains, creating docking sites for STAT3 and STAT5B transcription factors. Related references: PMID:9188471 PMID:8999038
- Additionally, the type II receptor recruits SHC1 via OSMR, leading to GRB2 association and activation of the Ras/Raf/MAPK signaling cascade. Related references: PMID:11016927
- OSM regulates diverse biological processes, including cell proliferation, differentiation, and cytokine production, and plays a role in inflammation, hematopoiesis, and tissue remodeling. Related references: PMID:10997905 PMID:1542792 PMID:1542793
- The protein is synthesized as a precursor with a signal peptide and is secreted, where it functions as a mitogen and growth regulator.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the mature Oncostatin M protein (amino acids 26–252), which lacks the signal peptide, likely enabling recognition of the secreted, biologically active form.
- For Western blot (WB), the predicted molecular weight of the unmodified monomer is approximately 28 kDa; however, glycosylation may shift the band, so consider using positive controls expressing OSM.
- This polyclonal antibody is suitable for detecting OSM in human, mouse, and rat samples, but cross-reactivity should be validated for each specific application.
- For IF/ICC, ensure appropriate permeabilization and fixation conditions, as secreted cytokines may require optimized protocols.
- In ELISA, use as a capture or detection antibody after confirming pairing compatibility with available OSM standards.
