- Nucleoredoxin (NXN), also known as NRX, RRS2, or TRG-4, is a 48 kDa redox-active protein belonging to the thioredoxin family. It contains a conserved thioredoxin domain and functions as an NAD-dependent oxidoreductase.
- Acts as a redox-dependent negative regulator of the canonical Wnt signaling pathway. It likely prevents the ubiquitination of Dishevelled-3 (DVL3) by the BCR(KLHL12) E3 ubiquitin ligase complex, thereby stabilizing DVL3 and inhibiting downstream β-catenin signaling.
- May also serve as a transcriptional regulator and a regulator of protein phosphatase 2A (PP2A), as suggested by similarity to other nucleoredoxins.
- Subcellularly, nucleoredoxin is found in both the cytoplasm (cytosol) and the nucleus, consistent with its roles in signal transduction and transcriptional regulation.
