Biological Background: NUR77 (NR4A1) Function and Localization
- Orphan nuclear receptor that binds the NGFI-B response element (NBRE) 5'-AAAGGTCA-3' and 9-cis-retinoic acid outside its ligand-binding domain. Related references: PMID:18690216, PMID:8121493, PMID:9315652
- Participates in energy homeostasis by sequestering the kinase STK11 in the nucleus, thereby attenuating cytoplasmic AMPK activation. Related references: PMID:22983157
- Regulates the inflammatory response in macrophages by repressing transcription of genes involved in the citric acid cycle (TCA).
- Inhibits NF-kappa-B signaling by binding to low-affinity NF-kappa-B binding sites, such as at the IL2 promoter. Related references: PMID:15466594
- In the cytosol, upon detection of bacterial LPS and mitochondrial DNA released during pyroptosis, promotes non-canonical NLRP3 inflammasome activation.
- May participate in liver regeneration and vascular response to injury.
- Predominantly nuclear, but also detected in cytoplasm, cytosol, and mitochondrion; expressed in fetal muscle, adult liver, brain, and thyroid.
- Subject to post-translational modifications including acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 200–300 of human NUR77, a region selected for its likely specificity and antigenicity; verify epitope accessibility in your assay.
- For western blotting, a band at approximately 64 kDa is expected; confirm molecular weight markers and optimize blocking conditions.
- In immunofluorescence/ICC, anticipate nuclear and cytoplasmic staining patterns; co-staining with organelle markers may help resolve localization.
- ELISA protocols should include appropriate controls (e.g., antigen-coated wells, negative sera) to ensure signal specificity.
