Biological Background: NTF3 Function and Localization
- Neurotrophin-3 (NTF3) is a secreted growth factor belonging to the neurotrophin family, which also includes NGF, BDNF, and NT-4/5.
- The NTF3 gene encodes a precursor protein that undergoes proteolytic cleavage at a pair of basic residues to generate the mature active form.
- Mature NTF3 forms a homodimer stabilized by disulfide bonds and is glycosylated, contributing to its stability and secretion.
- NTF3 plays a critical role in promoting the survival and differentiation of visceral and proprioceptive sensory neurons.
- It is expressed in brain and peripheral tissues, suggesting both central and peripheral neurotrophic functions.
- Alternative splicing of the NTF3 gene produces multiple isoforms, potentially regulating tissue-specific activity or localization.
- Keywords associated with NTF3 include growth factor, secreted, glycoprotein, disulfide bond, and alternative splicing, reflecting its biochemical properties and regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 19–257 of human NTF3, covering the mature neurotrophin domain after signal peptide cleavage. The resulting polyclonal antibody is expected to recognize both the precursor and mature forms.
- In Western blotting, consider that the mature NTF3 monomer after cleavage is predicted at ~13.6 kDa, but the glycosylated dimer may migrate at around 29 kDa under non-reducing conditions. Validate the observed band pattern in the relevant sample system.
- For immunohistochemistry (IHC-P), antigen retrieval may be required. Test in brain or peripheral nerve tissue sections where NTF3 expression is expected; strong neuronal staining is anticipated.
- This polyclonal antibody can be used in ELISA as a capture or detection reagent. For sandwich ELISA, pairing with a monoclonal antibody specific to a non-overlapping epitope may improve sensitivity.
- Cross-reactivity with mouse NTF3 is reported; however, sequence homology with other neurotrophins warrants evaluation of specificity in each application.
